PEG-MGF (PEGylated Mechano Growth Factor) is a splice variant of Insulin-like Growth Factor 1 (IGF-1) that has been chemically modified with polyethylene glycol (PEG) to extend its biological half-life from minutes to several hours. MGF is produced naturally in muscle tissue as an immediate response to mechanical stress such as exercise or injury, initiating the repair cascade before the liver-derived systemic IGF-1 response. Research has focused on PEG-MGF's ability to activate muscle satellite (stem) cells, which are quiescent myogenic precursor cells that reside between the sarcolemma and basal lamina of muscle fibers. Upon activation, these cells proliferate, differentiate, and fuse with existing myofibers or form new fibers. Studies have also explored PEG-MGF's neuroprotective effects, including its potential to protect motor neurons and promote neural stem cell proliferation. The PEGylation modification prevents rapid degradation by proteases and reduces renal clearance, making it particularly valuable for research protocols requiring sustained exposure to the growth factor.
