DSIP (Delta Sleep-Inducing Peptide) is a neuropeptide originally isolated in 1977 from the cerebral venous blood of rabbits during electrically induced slow-wave sleep. This amphiphilic nonapeptide (Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu) crosses the blood-brain barrier and has been researched for its ability to modulate sleep architecture, particularly by promoting delta (slow-wave) sleep without significantly altering REM sleep duration. DSIP's mechanism of action involves modulation of GABAergic, glutamatergic, and serotonergic neurotransmission, along with effects on the hypothalamic-pituitary-adrenal (HPA) axis. Research has demonstrated its potential to normalize disrupted sleep patterns, reduce ACTH and cortisol levels under stress conditions, modulate circadian rhythm phase shifts, and influence body temperature regulation. Additional research areas include pain perception modulation (through interaction with opioid signaling), alcohol and opioid withdrawal symptom management, oxidative stress reduction in neural tissue, and epilepsy research. DSIP has also been studied for its immunomodulatory effects, including enhancement of NK cell activity.
