Research overview
KPV is a tripeptide (Lys-Pro-Val) corresponding to the C-terminal sequence (residues 11-13) of alpha-melanocyte stimulating hormone (α-MSH). Despite its small size, KPV retains the potent anti-inflammatory activity of the parent hormone while lacking its melanogenic effects. Research has demonstrated that KPV exerts its anti-inflammatory properties through multiple mechanisms: direct inhibition of NF-κB nuclear translocation by preventing IκBα phosphorylation, reduction of pro-inflammatory cytokines (TNF-α, IL-1β, IL-6, IL-8), suppression of nitric oxide synthase (iNOS) expression, and modulation of the PGE2/COX-2 inflammatory axis. In preclinical inflammatory bowel disease models, KPV demonstrated significant reduction in colonic inflammation, mucosal damage, and disease activity indices. The peptide has also been studied in skin inflammation models (psoriasis, dermatitis), where it showed reduced keratinocyte inflammatory responses. Additionally, KPV exhibits antimicrobial properties against Staphylococcus aureus and Candida albicans, suggesting dual anti-inflammatory and antimicrobial functionality. Its small size and stability make it particularly amenable to topical and oral formulation development.
