Snap-8 (Acetyl Octapeptide-3, INCI: Acetyl Glutamyl Heptapeptide-1) is a synthetic octapeptide that mimics the N-terminal end of SNAP-25 (Synaptosomal-Associated Protein of 25 kDa), a critical component of the SNARE complex responsible for synaptic vesicle fusion and neurotransmitter release at the neuromuscular junction. By competing with native SNAP-25 for incorporation into the SNARE complex, Snap-8 destabilizes the complex and reduces the efficiency of acetylcholine release, thereby attenuating muscle contraction force. This mechanism is analogous to botulinum neurotoxin type A but operates through competitive inhibition rather than enzymatic cleavage. Research has focused on its application in cosmetic and dermatological models, with studies demonstrating dose-dependent reduction in muscle contraction depth in skin explant models. It is one of the most widely studied cosmeceutical peptides, with applications in anti-aging skincare research, neuromuscular junction biology, and topical peptide delivery optimization studies.
